NP-12, a thiadiazolidinone, is a selective (non ATP-competitive) inhibitor of glycogen synthase kinase 3ß (GSK3ß). In addition to be involved in the processes of cellular signaling of insulin and IGF-1, GSK3ß is highly expressed in the brain, with evidence that it is involved in the hyperphosphorylation of tau protein and increased amyloid beta production, which respectively form the hallmark neurofibrillary tangles and plaques in Alzheimer's disease.
